I'm unable to play games or load the BIOS from my USB hard drive, however I'm able to launch the emulator itself from the hard drive.
It says:
"Error opening directory USB:/WiiSX/isos"
"Error opening directory USB:/WiiSX/BIOS"
I have yet to try putting the content on my SD card because the card is very small, but I will try it and post back.
*Update* Both the BIOS and the game (Chrono Trigger) are successfully loaded from an SD card.

I have this same problem hard drive is formatted FAT32, every other app has no problem accessing the harddrive. WiiSX itself is launched from the USB drive and yet when it cant access the bios from the same harddrive the emulator was launched from. It also can't detect the drive to load ISOs either. I have no problems running anything from an SD card the problem is I dont have enough SD cards to store all the games I want to play so I would like to be able to use my external harddrive to run everything. Can somebody please help me with this?

drive is also unpartitioned I am going to try formatting it NTFS then FAT32 again just to make completely sure its unpartitioned and wiped

Have you tried the USB2 mod for IOS 202 or IOS 58?
